摘要: Frequenttrafficaccidentsconstituteamajordangertohumanbeings.Theaccident-pronedriverwhohasthestablephysiological,psychological,andbehavioralcharacteristicsisoneofthemostprominentcausesoftrafficaccidents.Theinternallinkbetweentheindividualcharacteristicsandtheaccidentpronenesshasbeenadifficultpointintheaccidentpreventionresearch.Theauthorsselectedaccident-pronedriversascasesandsafedriversascontrols(case-controlgroup)from18,360driverswhowereenrolledfromthreepublictransportationincorporationsofChinausingareastratifiedsamplingmethod.Thecase-controlgroupswere1:1matched.Theauthorsperformedgenome-wideassociationstudy(GWAS)by179casesand179controlsusingtheU.S.AffymetrixGenome-WideHumanMappingSNP6.0Array.Theauthorsobservedthatthegenefrequenciesof34single-nucleotidepolymorphisms(SNPs)inthreeregionsofcaseswerehigherthanthoseinthecontrol(P<10~(–4)).Theauthorsthentestedtwoindependentreplicationsetsforstrongassociation6SNPsin349pairsofcase-controldriversusingtheU.S.ABI3730sequencingmethod.TheresultsindicatedthatSNPrs6069499withinlinkedCBLN4genearestronglyassociatedwithaccidentproneness(Pcombined=6.37×10~(-10)).AccordingtoCBLN4genemainlyinvolvedinadrenaldevelopmentandtheregulationofsecretion,theauthorsperformed12biochemicalparametersofthebloodusingradioimmunoassay.Thelevelsofdopamine(DA)andadrenocorticotropic(ACTH)hormoneshowedsignificantdifferencesbetweenaccidentpronedriversandsafedrivers(P_(DA)=0.03,P_(ACTH)=0.01).Itissuggestedthattheaccident-pronedriversmayhavetheidiosyncrasyofsusceptibility. ...
