G~(57)mutationintheS-x-Emotif.Theenamelmicrostructureof4-week-oldmicewasexaminedbyscanningelectronmicroscopy.Theteethof6-day-oldmicewerecharacterizedbyhistologyandimmunohistochemistry.Theproteinlysatesofthefirstlowermolarsof4-day-oldmicewereanalyzedbyWesternimmunoblottingusingantibodiesagainstENAM,ameloblastinandamelogenin.ENAM~(Rgsc514)heterozygotesshowedadisorganizedenamelmicrostructure,whilethehomozygoteshadnoenamelonthedentinsurface.TheN-terminalfragmentsofENAMintheheterozygotesweredetainedintheameloblastsandlocalizedinthemineralizationfrontofenamelmatrix,whilethoseintheWTmiceweresecretedoutofameloblastsanddistributedevenlyintheouter1/2ofenamelmatrix.Surprisingly,the~15kDaC-terminalfragmentsofameloblastinwerenotdetectedinthemolarlysatesofthehomozygotes.TheseresultssuggestthatthephosphorylationofSer~(55)maybeanessentialposttranslationalmodificationofENAMandisrequiredfortheinteractionwithotherenamelmatrixmoleculessuchasameloblastininmediatingthestructuralorganizationofenamelmatrixandprotein-mineralinteractionsduringenamelformation.">
